Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_020778.5(ALPK3):c.2747T>C (p.Leu916Pro), citing LabCorp Variant Classification Summary - May 2015: Variant summary: ALPK3 c.2747T>C (p.Leu916Pro) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. The variant allele was found at a frequency of 0.00023 in 243890 control chromosomes. This frequency is not significantly higher than estimated for disease-causing variants in ALPK3, allowing no conclusion about variant significance. c.2747T>C has been observed in at least one individual affected with Dilated Cardiomyopathy. This individual had co-occurrences in three other genes associated with cardiac phenotypes, including a pathogenic variant in the SCN5A gene (example: Burstein_2021). This report does not provide unequivocal conclusions about association of the ALPK3 variant with Cardiomyopathy. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
