NM_007254.4(PNKP):c.178C>G (p.Arg60Gly) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R60G variant (also known as c.178C>G), located in coding exon 2 of the PNKP gene, results from a C to G substitution at nucleotide position 178. The arginine at codon 60 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
