Likely pathogenic — the classification assigned by GeneDx to NM_004830.4(MED23):c.599T>G (p.Leu200Ter), citing GeneDx Variant Classification (06012015): The L200X variant in the MED23 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The L200X variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). We interpret L200X as a likely pathogenic variant.
