This variant lies in the AIFM1 gene (transcript NM_004208.4) at coding-DNA position 469, where G is replaced by T; at the protein level this means replaces alanine at residue 157 with serine — a missense variant. Submitter rationale: The A157S variant in the AIFM1 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The A157S variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The A157S variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret A157S as a likely pathogenic variant.
