This variant lies in the NPR2 gene (transcript NM_003995.4) at coding-DNA position 2833, where C is replaced by G; at the protein level this means replaces arginine at residue 945 with glycine — a missense variant. Submitter rationale: The R945G variant in the NPR2 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The R945G variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The R945G variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position where amino acids with similar properties to Arginine are tolerated across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret R945G as a variant of uncertain significance.
