Uncertain significance — the classification assigned by GeneDx to NM_001852.4(COL9A2):c.1665G>A (p.Met555Ile), citing GeneDx Variant Classification (06012015): A variant of uncertain significance has been identified in the COL9A2 gene. The M555I variant has not been published as pathogenic or been reported as benign to our knowledge. This variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). In silico analysis predicts this variant is probably damaging to the protein structure/function. However, the M555I variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. This substitution occurs at a position where amino acids with similar properties to methionine are tolerated across species and where isoleucine is present as the wild type in at least one species.
