NM_052854.4(CREB3L1):c.46G>T (p.Gly16Ter) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the CREB3L1 gene (transcript NM_052854.4) at coding-DNA position 46, where G is replaced by T; at the protein level this means converts the codon for glycine at residue 16 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The G16X variant in the CREB3L1 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The G16X variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). We interpret G16X as a variant of uncertain significance.
