NM_020745.4(AARS2):c.424G>A (p.Glu142Lys) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the AARS2 gene (transcript NM_020745.4) at coding-DNA position 424, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 142 with lysine — a missense variant. Submitter rationale: The E142K variant in the AARS2 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The E142K variant is not observed in large population cohorts (Lek et al., 2016). The E142K variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. In-silico analyses, including protein predictors and evolutionary conservation, support a deleterious effect. We interpret E142K as a variant of uncertain significance.
