NM_000089.4(COL1A2):c.607G>C (p.Ala203Pro) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the COL1A2 gene (transcript NM_000089.4) at coding-DNA position 607, where G is replaced by C; at the protein level this means replaces alanine at residue 203 with proline — a missense variant. Submitter rationale: The A203P variant in the COL1A2 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The A203P variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The A203P variant is a semi-conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties. This substitution occurs at a position that is conserved across species. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret A203P as a variant of uncertain significance.
