This variant lies in the FAM20C gene (transcript NM_020223.4) at coding-DNA position 739, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 247 with lysine — a missense variant. Submitter rationale: The E247K variant in the FAM20C gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The E247K variant is observed in 34/16330 (0.2%) alleles from individuals of South Asian background, in large population cohorts (Lek et al., 2016). The E247K variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is not conserved. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret E247K as a variant of uncertain significance.
