NM_182894.3(VSX2):c.91G>A (p.Gly31Arg) was classified as Likely pathogenic by GeneDx, citing GeneDx Variant Classification (06012015): The G31R variant in the VSX2 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The G31R variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved across species, and in silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret G31R as a likely pathogenic variant.
