This variant lies in the ADSL gene (transcript NM_000026.4) at coding-DNA position 805, where A is replaced by G; at the protein level this means replaces isoleucine at residue 269 with valine — a missense variant. Submitter rationale: A variant of uncertain significance has been identified in the ADSL gene. The I269V variant has not been published as a pathogenic variant, nor has it been reported as a benign variant to our knowledge. The I269V variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). This substitution occurs at a position that is conserved across species. Missense variants in nearby residues (D268H/N) have been reported in Human Gene Mutation Database in association with ADSL deficiency (Stenson et al., 2014), supporting the functional importance of this region of the protein.
