Submitter rationale: The V410I variant, present in an alternate transcript of the KDM6A gene, has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The V410I variant is observed in 2/12636 (0.0016%) alleles in large population cohorts (Lek et al., 2016). The V410I variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. In-silico analyses, including protein predictors and evolutionary conservation, support that this variant does not alter protein structure/function. We interpret V410I as a variant of uncertain significance.
