NM_000426.4(LAMA2):c.6605G>T (p.Gly2202Val) was classified as Likely pathogenic by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the LAMA2 gene (transcript NM_000426.4) at coding-DNA position 6605, where G is replaced by T; at the protein level this means replaces glycine at residue 2202 with valine — a missense variant. Submitter rationale: The G2202V variant in the LAMA2 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The G2202V variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The G2202V variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. However, this substitution occurs at a position that is conserved across species and in silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret G2202V as a likely pathogenic variant.
