This variant lies in the PIEZO2 gene (transcript NM_001378183.1) at coding-DNA position 1535, where G is replaced by A; at the protein level this means replaces serine at residue 512 with asparagine — a missense variant. Submitter rationale: The S512N variant in the PIEZO2 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The S512N variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The S512N variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. This substitution occurs at a position that is conserved across species, and in silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret S512N as a variant of uncertain significance.
