Uncertain significance — the classification assigned by GeneDx to NM_177438.3(DICER1):c.128C>G (p.Thr43Arg), citing GeneDx Variant Classification (06012015): The T43R variant in the DICER1 gene has not, to our knowledge, been published in the literature as pathogenic or benign. This variant was not observed in large population cohorts (NHLBI Exome Sequencing Project, The 1000 Genomes Consortium 2015, Lek et al., 2016). Since Threonine and Arginine differ in some properties, this is considered a semi-conservative amino acid substitution. This substitution occurs at a position that is conserved across species and in silico analyses predict that this variant is probably damaging to protein structure and function. Based on currently available evidence, we consider T43R to be a variant of uncertain significance.
