NM_000540.3(RYR1):c.6884T>C (p.Leu2295Pro) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the RYR1 gene (transcript NM_000540.3) at coding-DNA position 6884, where T is replaced by C; at the protein level this means replaces leucine at residue 2295 with proline — a missense variant. Submitter rationale: The L2295P variant in the RYR1 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The L2295P variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The L2295P variant is a semi-conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties. This substitution occurs at a position that is conserved across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret L2295P as a variant of uncertain significance.
