This variant lies in the CHRNB2 gene (transcript NM_000748.3) at coding-DNA position 632, where G is replaced by A; at the protein level this means replaces arginine at residue 211 with glutamine — a missense variant. Submitter rationale: The R211Q variant in the CHRNB2 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The R211Q variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The R211Q variant is a semi-conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties. This substitution occurs at a position where amino acids with similar properties to Arginine are tolerated across species. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret R211Q as a variant of uncertain significance.
