NM_001371623.1(TCOF1):c.1923_1924dup (p.Lys642fs) was classified as Pathogenic by GeneDx, citing GeneDx Variant Classification (06012015): The c.1923_1924dupCA variant in the TCOF1 gene causes a frameshift starting with codon Lysine 642, changes this amino acid to a Threonine residue and creates a premature Stop codon at position 70 of the new reading frame, denoted p.Lys642ThrfsX70. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. Although this variant has not been previously reported to our knowledge, we consider it to be pathogenic.
