Submitter rationale: The P346A variant in the PAX6 gene has been previously reported as a heterozygous finding in an individual with hyperopia and cataract (Hewitt et al., 2007). This variant is observed in 2/15604 (0.013%) alleles from individuals of South Asian background in the ExAC dataset (Lek et al., 2016). The P346A variant is a semi-conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties. This substitution occurs at a position that is conserved in mammals.
