NM_000384.3(APOB):c.154T>C (p.Tyr52His) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y52H variant (also known as c.154T>C), located in coding exon 3 of the APOB gene, results from a T to C substitution at nucleotide position 154. The tyrosine at codon 52 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
