NM_000257.4(MYH7):c.953C>A (p.Thr318Asn) was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015. This variant lies in the MYH7 gene (transcript NM_000257.4) at coding-DNA position 953, where C is replaced by A; at the protein level this means replaces threonine at residue 318 with asparagine — a missense variant. Submitter rationale: Variant summary: MYH7 c.953C>A (p.Thr318Asn) results in a non-conservative amino acid change located in the Myosin head, motor domain (IPR001609) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251392 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.953C>A has been reported in the literature in individuals affected with Hypertrophic Cardiomyopathy (Ho_2018). This report does not provide unequivocal conclusions about association of the variant with Cardiomyopathy. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication have been ascertained in the context of this evaluation (PMID: 30297972). No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as uncertain significance.
