Pathogenic for Aniridia 1; Irido-corneo-trabecular dysgenesis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001368894.2(PAX6):c.836G>A (p.Trp279Ter), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the PAX6 gene (transcript NM_001368894.2) at coding-DNA position 836, where G is replaced by A; at the protein level this means converts the codon for tryptophan at residue 279 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This sequence change creates a premature translational stop signal (p.Trp265*) in the PAX6 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in PAX6 are known to be pathogenic (PMID: 12634864). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with aniridia (PMID: 32360764). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 431004). For these reasons, this variant has been classified as Pathogenic.
