This variant lies in the L1CAM gene (transcript NM_001278116.2) at coding-DNA position 1485, where C is replaced by G; at the protein level this means converts the codon for tyrosine at residue 495 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The Y495X nonsense variant in the L1CAM gene is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The Y495X variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). Although this pathogenic variant has not been reported previously to our knowledge, its presence is consistent with the diagnosis of X-linked hydrocephalus
