This variant lies in the EGR2 gene (transcript NM_000399.5) at coding-DNA position 308, where C is replaced by T; at the protein level this means replaces proline at residue 103 with leucine — a missense variant. Submitter rationale: The P103L variant in the EGR2 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The P103L variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The P103L variant is a semi-conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties. This substitution occurs at a position that is not conserved. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret P103L as a variant of uncertain significance.
