Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000302.4(PLOD1):c.1780G>A (p.Glu594Lys), citing LabCorp Variant Classification Summary - May 2015: Variant summary: PLOD1 c.1780G>A (p.Glu594Lys) results in a conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 8e-06 in 250932 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1780G>A in individuals affected with Ehlers-Danlos syndrome, kyphoscoliotic type 1 and no experimental evidence demonstrating its impact on protein function have been reported.
