This variant lies in the SALL4 gene (transcript NM_020436.5) at coding-DNA position 1019, where G is replaced by A; at the protein level this means replaces glycine at residue 340 with aspartic acid — a missense variant. Submitter rationale: TThe G340D variant in the SALL4 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The G340D variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016). The G340D variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. In-silico analyses, including protein predictors and evolutionary conservation, support a deleterious effect. We interpret G340D as a variant of uncertain significance.
