NM_001510.4(GRID2):c.899G>A (p.Arg300His) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the GRID2 gene (transcript NM_001510.4) at coding-DNA position 899, where G is replaced by A; at the protein level this means replaces arginine at residue 300 with histidine — a missense variant. Submitter rationale: The R300H variant in the GRID2 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The R300H variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The R300H variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. This substitution occurs at a position where amino acids with similar properties to Arginine are tolerated across species. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret R300H as a variant of uncertain significance.
