Pathogenic — the classification assigned by GeneDx to NM_000388.4(CASR):c.3010del (p.Ser1004fs), citing GeneDx Variant Classification (06012015): The c.3010delA deletion in the CASR gene causes a frameshift starting with codon Serine 1004, changes this amino acid to an Alanine residue and creates a premature Stop codon at position 4 of the new reading frame, denoted p.Ser1004AlafsX4. This variant is predicted to cause loss of normal protein function through protein truncation as the last 75 amino acids are replaced with three aberrant amino acids. Although c.3010delA has not been previously reported to our knowledge, it is expected to be a pathogenic variant.
