Submitter rationale: The c.159_162delGAGT deletion in the PAX6 gene causes a frameshift starting with codon Serine 54, changes this amino acid to a Lysine residue and creates a premature Stop codon at position 24 of the new reading frame, denoted p.Ser54LysfsX24. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. Although this deletion has not been previously reported to our knowledge, we consider it to be a pathogenic variant.
