This variant lies in the EDA gene (transcript NM_001399.5) at coding-DNA position 922, where G is replaced by T; at the protein level this means converts the codon for glutamic acid at residue 308 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The E308X nonsense variant in the EDA1 gene is predicted to cause loss of normal protein function through protein truncation as the last 84 residues of the protein are lost. E308X was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. Although this variant has not been reported previously to our knowledge, we interpret it to be pathogenic.
