Uncertain significance — the classification assigned by GeneDx to NM_001347721.2(DYRK1A):c.1205G>A (p.Gly402Glu), citing GeneDx Variant Classification (06012015). This variant lies in the DYRK1A gene (transcript NM_001347721.2) at coding-DNA position 1205, where G is replaced by A; at the protein level this means replaces glycine at residue 402 with glutamic acid — a missense variant. Submitter rationale: The G411E variant in the DYRK1A gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The G411E variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The G411E variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret G411E as a variant of uncertain significance.
