This variant lies in the PCNT gene (transcript NM_006031.6) at coding-DNA position 934, where C is replaced by T; at the protein level this means replaces arginine at residue 312 with tryptophan — a missense variant. Submitter rationale: A variant of uncertain significance has been identified in the PCNT gene. The R312W variant has not been published as a pathogenic variant, nor has it been reported as a benign variant to our knowledge. The R312W variant is observed in 7/36,328 (0.02%) alleles from individuals of European background (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The R312W variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved in mammals.
