This variant lies in the MED13L gene (transcript NM_015335.5) at coding-DNA position 6073, duplicating one base; at the protein level this means shifts the reading frame starting at methionine residue 2025, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The c.6073dupA variant in the MED13L gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.6073dupA variant causes a frameshift starting with codon Methionine 2025, changes this amino acid to an Asparagine residue, and creates a premature Stop codon at position 4 of the new reading frame, denoted p.Met2025AsnfsX4. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.6073dupA variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). We interpret c.6073dupA as a pathogenic variant.
