This variant lies in the COL4A3 gene (transcript NM_000091.5) at coding-DNA position 4018, where G is replaced by C; at the protein level this means replaces glycine at residue 1340 with arginine — a missense variant. Submitter rationale: The variant is not observed in the gnomAD v4.1.0 dataset. Predicted Consequence/Location: Missense variant In silico tool predictions suggest damaging effect of the variant on gene or gene product [REVEL: 1.00 (>=0.6, sensitivity 0.68 and specificity 0.92)]. A different missense change at the same codon (p.Gly1340Glu) has been reported to be associated with COL4A3-related disorder (ClinVar ID: VCV000438698).
