NM_024514.5(CYP2R1):c.1126C>T (p.Pro376Ser) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015): The P376S variant in the CYP2R1 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The P376S variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The P376S variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret P376S as a variant of uncertain significance.
