Uncertain significance for Spondyloepimetaphyseal dysplasia, Isidor-Toutain type — the classification assigned by 3billion to NM_000977.4(RPL13):c.504_507dup (p.Thr170fs), citing ACMG Guidelines, 2015: The variant is not observed in the gnomAD v4.0.0 dataset. Predicted Consequence/Location: Frameshift: predicted to result in a loss or disruption of normal protein function through protein truncation. Multiple pathogenic variants are reported near the predicted truncated region and is predicted to have a dominant negative effect on protein function (PMID: 32916022). However, evidence are insufficient at this time. Therefore, this variant is classified as VUS according to the recommendation of ACMG/AMP guideline.
