Uncertain significance for Developmental and epileptic encephalopathy, 19 — the classification assigned by 3billion to NM_001127644.2(GABRA1):c.448G>A (p.Glu150Lys), citing ACMG Guidelines, 2015. This variant lies in the GABRA1 gene (transcript NM_001127644.2) at coding-DNA position 448, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 150 with lysine — a missense variant. Submitter rationale: The variant is not observed in the gnomAD v4.1.0 dataset. Predicted Consequence/Location: Missense variant. Missense changes are a common disease-causing mechanism. In silico tool predictions suggest damaging effect of the variant on gene or gene product [3Cnet: 0.84 (> 0.75, sensitivity 0.96 and precision 0.92)]. The same nucleotide change resulting in the same amino acid change has been previously reported to be associated with GABRA1-related disorder (PMID: 36584483). However, the evidence of pathogenicity is insufficient at this time. Therefore, this variant is classified as VUS according to the recommendation of ACMG/AMP guideline.
