Uncertain significance — the classification assigned by GeneDx to NM_018972.4(GDAP1):c.536C>A (p.Pro179Gln), citing GeneDx Variant Classification (06012015). This variant lies in the GDAP1 gene (transcript NM_018972.4) at coding-DNA position 536, where C is replaced by A; at the protein level this means replaces proline at residue 179 with glutamine — a missense variant. Submitter rationale: The P179Q variant in the GDAP1 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The P179Q variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The P179Q variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret P179Q as a variant of uncertain significance.
