This variant lies in the MEF2C gene (transcript NM_002397.5) at coding-DNA position 4, where G is replaced by A; at the protein level this means replaces glycine at residue 2 with arginine — a missense variant. Submitter rationale: The variant is not observed in the gnomAD v4.1.0 dataset. Predicted Consequence/Location: Missense variant In silico tool predictions suggest damaging effect of the variant on gene or gene product [REVEL: 0.83 (>=0.6, sensitivity 0.68 and specificity 0.92); 3Cnet: 0.99 (>=0.6, sensitivity 0.72 and precision 0.9)]. The different nucleotide change resulting in the same amino acid change has been previously reported to be associated with MEF2C-related disorder(PMID: 29655203). However, the evidence of pathogenicity is insufficient at this time. Therefore, this variant is classified as VUS according to the recommendation of ACMG/AMP guideline.
