Submitter rationale: CFH p.Trp134Arg (c.400T>C) is a missense variant that changes the amino acid at residue 134 from Tryptophan to Arginine. This variant has been observed in at least one proband affected with atypical hemolytic-uremic syndrome (PMID:23431077;21717289). At least one functional study has demonstrated a substantial alteration in protein function relative to the wild-type (PMID:34189567;36846022). It is absent or not present at a significant frequency in gnomAD. In silico models agree that this variant is possibly or probably damaging.
