This variant lies in the CFH gene (transcript NM_000186.4) at coding-DNA position 1422, deleting one base; at the protein level this means shifts the reading frame starting at lysine residue 474, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: CFH p.Lys474AsnfsTer6 (c.1422del) is a frameshift variant that results in the production of a truncated protein which is predicted to undergo nonsense-mediated mRNA decay. This variant has been observed in at least one proband affected with atypical hemolytic-uremic syndrome (PMID:11158219;33841858). The variant was found to segregate with disease in at least one affected family (PMID:11158219). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify CFH p.Lys474AsnfsTer6 (c.1422del) as a pathogenic variant.
