Submitter rationale: CD46 c.947-1G>C is a canonical splice variant located in the acceptor splice region in intron 8. It is predicted to affect mRNA splicing, leading to a deleterious effect on the CD46 protein. This variant has been observed in at least one proband affected with atypical hemolytic-uremic syndrome (PMID:37744338). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify CD46 c.947-1G>C as a pathogenic variant.
