NM_000051.3(ATM):c.8851-?_*3591dup3912 was classified as Uncertain significance for Ataxia-telangiectasia syndrome by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross duplication of the genomic region encompassing exons 62-63 of the ATM gene. The 5' boundary is likely confined to intron 61. The 3' end of this event is unknown as it extends beyond the assayed region for this gene and therefore may encompass additional genes. A similar duplication involving exons 62 and 63 (referred to as DupEx64+65) has been reported in an individual affected with ataxia-telangiectasia who also carried a pathogenic nonsense variant in ATM (PMID: 25122203). However, it was not determined whether these variants are on the same or opposite chromosomes. Experimental studies and prediction algorithms are not available for this variant, and the functional significance of the duplicated exons is currently unknown. In summary, the genomic location of this duplication is unknown and the impact of this variant on ATM protein function has not been established. Therefore, it has been classified as a Variant of Uncertain Significance.
