This variant lies in the NF1 gene (transcript NM_001042492.3) at the canonical splice acceptor site of the intron immediately before coding-DNA position 7063, where G is replaced by T; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The c.7000-1G>T intronic pathogenic mutation results from a G to T substitution one nucleotide upstream from coding exon 47 of the NF1 gene. Another pathogenic mutation at the same nucleotide position (c.7000-1G>A) has been described in a proband with NF1 (Sabbagh A et al. Hum. Mutat. 2013 Nov; 34(11):1510-8).
