This variant lies in the CFI gene (transcript NM_000204.5) at coding-DNA position 1582, where C is replaced by A; at the protein level this means replaces proline at residue 528 with threonine — a missense variant. Submitter rationale: CFI p.Pro528Thr (c.1582C>A) is a missense variant that changes the amino acid at residue 528 from Proline to Threonine. To our knowledge, this variant has not been reported in patients affected with CFI-related disorders in the published literature. At least one functional study has demonstrated a substantial alteration in protein function relative to the wild-type (PMID:38852887). It is absent or not present at a significant frequency in gnomAD. In silico models agree that this variant is possibly or probably damaging. In conclusion, we classify CFI p.Pro528Thr (c.1582C>A) as a likely pathogenic, low penetrance variant.
