Uncertain significance for CFI-related disorder — the classification assigned by Genomenon, Inc, Genomenon, Inc to NM_000204.5(CFI):c.715A>C (p.Lys239Gln), citing Genomenon Sequence Variant Interpretation Standards - Updated. This variant lies in the CFI gene (transcript NM_000204.5) at coding-DNA position 715, where A is replaced by C; at the protein level this means replaces lysine at residue 239 with glutamine — a missense variant. Submitter rationale: CFI p.Lys239Gln (c.715A>C) is a missense variant that changes the amino acid at residue 239 from Lysine to Glutamine. To our knowledge, this variant has not been reported in patients affected with CFI-related disorders in the published literature. Functional studies have been reported; however, the significance of the findings remain unclear (20044478). The variant is located in a mutational hotspot. It is absent or not present at a significant frequency in gnomAD. In silico models agree that this variant is not damaging. In conclusion, we classify CFI p.Lys239Gln (c.715A>C) as a variant of unknown significance.
