Likely pathogenic, low penetrance for CFI-related disorder — the classification assigned by Genomenon, Inc, Genomenon, Inc to NM_000204.5(CFI):c.930C>A (p.Asp310Glu), citing Genomenon Sequence Variant Interpretation Standards - Updated: CFI p.Asp310Glu (c.930C>A) is a missense variant that changes the amino acid at residue 310 from Aspartic acid to Glutamic acid. To our knowledge, this variant has not been reported in patients affected with CFI-related disorders in the published literature. At least one functional study has demonstrated a substantial alteration in protein function relative to the wild-type (PMID:32510551). It is absent or not present at a significant frequency in gnomAD. In silico models agree that this variant is not damaging. In conclusion, we classify CFI p.Asp310Glu (c.930C>A) as a likely pathogenic, low penetrance variant.
