Submitter rationale: CFI p.Cys106Arg (c.316T>C) is a missense variant that changes the amino acid at residue 106 from Cysteine to Arginine. To our knowledge, this variant has not been reported in patients affected with CFI-related disorders in the published literature. At least one functional study has demonstrated a substantial alteration in protein function relative to the wild-type (PMID:32510551). It is absent or not present at a significant frequency in gnomAD. In silico models agree that this variant is possibly or probably damaging.
