NM_003647.3(DGKE):c.501C>G (p.Cys167Trp) was classified as Likely pathogenic, low penetrance for Atypical hemolytic-uremic syndrome by Genomenon, Inc, Genomenon, Inc, citing Genomenon Sequence Variant Interpretation Standards - Updated. This variant lies in the DGKE gene (transcript NM_003647.3) at coding-DNA position 501, where C is replaced by G; at the protein level this means replaces cysteine at residue 167 with tryptophan — a missense variant. Submitter rationale: DGKE p.Cys167Trp (c.501C>G) is a missense variant that changes the amino acid at residue 167 from Cysteine to Tryptophan. This variant has been observed in at least one proband affected with atypical hemolytic-uremic syndrome (PMID:25443527). At least one functional study has demonstrated a substantial alteration in protein function relative to the wild-type (PMID:36113832). It is absent or not present at a significant frequency in gnomAD. In silico models agree that this variant is possibly or probably damaging. In conclusion, we classify DGKE p.Cys167Trp (c.501C>G) as a likely pathogenic, low penetrance variant.
